Desmosine is prepared according to the method of Starcher et al. from hydrolysed neck ligament by solvent extraction and chromatography on Dowex 50.
NOTE: Extremely hygroscopic and must be stored over a desiccant such as silica gel.
Desmosine is a tetrafunctional, pyridinium ring-containing amino acids involved in elastin cross-linking. A desmosine cross-link is formed from three allysyl side chains plus one unaltered lysyl side chain from the same or neighbouring polypeptides.
Desmosine is responsible for the cross-linking of elastin leading to the formation of insoluble fibers. Urinary excretion of desmosine is a useful indicator of elastin breakdown resulting from increased elastase activity. It also acts as a biomarker of elastin degradation in chronic obstructive.
