Metabolism of the TMB substrate by HRP creates a soluble bluish reaction product that may be quantified by measuring absorbance at 370 or 665 nm. Reaction can be stopped by addition of 1 volume of 1N hydrocloric or sulfurc acid to yield a yellow reaction product that can be measured by absorbance at 450 nm (within one hour).
