Description: Methotrexate is a cell cycle arresting agent with varying effects. Methotrexate has been reported to arrest the cell cycle in late G1/S thus leading to the inhibition of the synthesis of DNA, RNA, thymidylates, and proteins. The main mechanism of action is reported to involve the inhibition of enzymes involved in purine metabolism which leads to the accumulation of adenosine or the suppression of intercellular adhesion molecule expression by T cells. Additionally, this compound has been observed to inhibit DHFR.
Methotrexate is used for chemotherapy either alone or in combination with other agents. It is effective for the treatment of a number of cancers including: breast, head and neck, leukemia, lymphoma, lung, osteosarcoma, bladder, and trophoblastic neoplasms. It is also used in treatment of autoimmune diseases, ectopic pregnancy, and for the induction of medical abortions. Brefeldin A is a fungal metabolite which is a macrocyclic lactone exhibiting a wide range of antibiotic activity. Produced by Penicillium brefeldianum. Blocks binding of the cytosolic coat protein b-COP and ARF to Golgi membranes mediated by protein G. Also blocks protein transportation into post-Golgi compartments. It activates the sphingomyelin cycle. Brefeldin A mediated apoptosis has been observed in human tumor cells.
Brefeldin A reversibly inhibits the intracellular translocation of proteins in eukaryotes, e.g., during transport of proteins to the cell surface for secretion or expression. It has been reported to block the response of cultured cells to cholera toxin. In HepG2 cells, BFA induces two blocks in the secretory pathway; one at the level of the endoplasmic reticulum-Golgi juncture and the other in the trans-Golgi network. Glutathione is the major low molecular weight thiol compound of the living plant or animal cell. It is a tripeptide with a gamma peptide linkage between the amine group of cysteine (which is attached by normal peptide linkage to a glycine) and the carboxyl group of the glutamate side-chain. It is an antioxidant, preventing damage to important cellular components caused by reactive oxygen species such as free radicals and peroxides. The sulfhydryl (thiol) group (SH) of cysteine serves as a proton donor and is responsible for the biological activity of glutathione.
Glutathione suppresses human immunodeficiency virus expression in chronically infected monocytic cells. It is a useful tripeptide involved in many aspects of metabolism, including transport of g-glutanyl amino acids and reductive cleavage of disulfide bonds.
Endogenous antioxidant that plays a major role in reducing reactive oxygen species formed during cellular metabolism and the respiratory burst.
